The Kansas City Athletics selected Jackson on June 7 in the 1966 Major League Baseball Draft. He was the second overall pick and spent one year in the minor league. He made his professional debut as the center fielder in the season opener on June 24 at Bethel Park in Eugene, Oregon. In 1966, he played for two class A teams with the Broncs only for 12 games, then 56 games with Modesto in the California League, where he hit 21 homers. His debut in the major leagues was a doubleheader in Kansas City as part of the athletics.

The Athletics moved west to be based in Oakland in 1968; Jackson hit 47 home runs in 1969 and was briefly ahead of the peace of Babe Ruth when he set the previous record of 60 in 1927. He played with the Athletics until 1975 before playing with the Baltimore Orioles for only one season. He helped athletics win several pennants and was named the MVP for the 1973 season.
